What is tirzepatide?

Tizepamide is a linearly synthesized peptide containing 39 amino acids partially conjugated with C20 fatty acids. Its protein sequence is based on endogenous GIP sequences, and its pharmacological effects on GLP-1 receptors are equivalent to endogenous GIP; However, the half-life of Tizepamide is relatively long and weekly administration is allowed. Texaparide was approved by FDA on May 13, 2022, with the trade name of MOUNTJARO, for the treatment of adult type 2 diabetes, making it the first and only GIP and GLP-1 receptor agonist for this indication. Later, on November 8, 2023, it was approved under a different brand name ZEPBOUND for long-term weight management in adults with at least one type of weight related obesity or overweight. On September 15, 2022, ticalopatide was also approved by the European Commission. On May 21, 2024, Tirzepatide was approved by the China National Medical Products Administration for the treatment of adult type 2 diabetes,with the brand name MOUNTJARO.

Glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) are hormones involved in blood-sugar control. After a person has eaten, these hormones are secreted by cells of the intestines, and in turn cause the secretion of insulin. Tirzepatide is a long-acting GIP-analogue that activates both the GLP-1 and GIP receptors, leading to improved blood-sugar control.
